Work History

Senior Research Scientist

Medytox, Inc
Suwon, Gyeonggi-do
09.2008 - Current
  • Design, perform and reporting of various experiments followed by trouble shooting and managing of projects was the major role.
  • Group work and managing of members within and outside of the team was the frequent job
  • Several immune libraries against specific antigens were constructed from where high affinity antibodies (at sub-nanomolar) were isolated via phage display technology.
  • Several folds of affinity of an antibody was enhanced by affinity maturation.
  • Highly purified and quality antigens such as recombinants of VEGF, TNF, PDGF etc was expressed from host cells
  • The vectors for recombinant protein in mammalian expression system transiently in ExpiCHO or stably in CHO-K1 were designed, constructed and effectively tested for high production.
  • With shorten time line, the stable cell line for various formats of antibodies in CHO-K1 (HORIZON) was produced in gram/L scale using FACS, VIPS and other techniques.
  • Highly efficient of RCB, WCB and and MCB for stable cell line were prepared.
  • Various in vitro and in vivo bio-assay for characterization of antibody functions, and quantification were developed.
  • Supervised junior technicians and scientists.
  • Wrote reports, reviews and summaries regarding the antibody projects.

Fellowship

Postdoctoral, EWHA Womans University
Seoul, Seoul
03.2006 - 08.2008
  • Maintained accurate records of research findings and provided statistical analysis of data results.
  • Contributed to and actively participated in research conception, design and execution to address defined problems.
  • Worked to improve collective knowledge of genetic engineering of bacteria for secondary metabolite production.
  • The biosynthesis of FK506, an immunosuppressant macrolide was further confirmed via generating of various mutant strains and isolation of intermediates after feeding of several starter units
  • The pathway was based on a comprehensive chemical, biochemical, and genetic interrogation of three FK506 gene clusters
  • Chemically difficult-to-synthesis intermediate for a ketolide, erythromycin, was produced by combinatorial biosynthesis in Saccharopolyspora erythraea that shorten and reduce the chemical synthesis steps.

Doctoral Researcher

Sunmoon University
Asan, ChungChungNamdo
03.2001 - 12.2005
  • Genetic Engineering: the functional characterization of a gene or a cluster of genes responsible for medicinally important small molecule (drugs) was achieved via deletion and complementation of gene form the corresponding gene deleted-strain
  • The gene function was further proven by expression of recombinant proteins in Streptomyces and E. Coli system
  • A door for genetic engineering for several antibiotics in practice such as tobramycin, kanamycin and gentamycin (aminoglycosides) or Angucyclines such as Sch 47554 and Sch 47555 were opened by identifying of the gene clusters from the corresponding strain by cosmid-library and screening of bio-synthetic gene clusters
  • A genomic analysis for secondary metabolites from Streptomyces peucetius ATCC 27952, a type of chemotherapy drug Doxorubicin producer, was successfully carried out
  • The identification and comparison of cytochrome P450 from S. Peucetius ATCC 27952 with other Streptomyces provided a tool for secondary metabolite modification
  • It was proven by a structural diversification of macrolactones by substrate‐flexible cytochrome P450 monooxygenases and identification of the role of erythromycin C-12 hydroxylase, EryK, as a substitute for PikC hydroxylase in pikromycin biosynthesis.
