Conducted research on yeast metabolic engineering projects focused on mixed-sugar utilization, glucose repression, and alternative carbon-source metabolism.
Designed gRNAs for CRISPR/Cas9-mediated gene deletion and analyzed mixed-sugar fermentation profiles to investigate candidate genes involved in glucose repression alleviation in Saccharomyces cerevisiae.
Engineered lactose-utilizing Saccharomyces cerevisiae strains through PCR, molecular cloning, plasmid construction, and yeast transformation of lactose-utilization genes.
Conducted flask-scale fermentation experiments using Saccharomyces cerevisiae and Kluyveromyces spp. under various single- and mixed-sugar conditions, with time course sampling and HPLC-based analysis of sugar consumption and fermentation metabolite profiles.
Applied adaptive laboratory evolution and phenotypic screening to select Kluyveromyces lactis strains with improved xylose utilization.
Attempted GFP-fusion protein analysis of MTH1 and mutant MTH1, including yeast protein extraction, BCA assay, SDS-PAGE, and anti-GFP Western blotting.
Addressed unexpected experimental results across research projects through repeated trials, comparison of cultivation conditions, and investigation of possible causes based on published studies.
